Here, we describe a method for isolating the stromal, the thylakoid membrane, and the thylakoid lumen subchloroplast fractions from Arabidopsis thaliana leaf material. All three fractions can be isolated sequentially from the same plant material in a single day preparation.

Genes encoding similar putative thylakoid cyclophilins with a unique insert of three amino acids NPV in their N-termini were found in chromosome 5 of both Arabidopsis and rice. TLP20 is suggested to be the major PPIase and protein folding catalyst in the thylakoid lumen of plant chloroplasts.

Figure legend: In the chloroplast's thylakoid membranes, electrons in a pigment-protein complex, called photosystem II (PS-II), excited by PS-II-captured photons, are shuttled to the next step in an electron-chain of proteins, the transported electrons replaced by PS-II-splitting (oxidation) of water molecules (H 2 O), releasing protons (H + ions) and oxygen into the thylakoid lumen.

(2011). Sodium fluoride was included in thylakoid isolation buffers for samples intended for BN-PAGE, whilst excluded from thylakoids used for spectroscopy analyses (see below).

The thylakoid lumen, a dynamic part of the photosynthetic process Research project The beautiful architecture of the thylakoid network is a distinguishing characteristic of the chloroplast of higher plants.

2017-10-23 The electron transport chain moves protons across the thylakoid membrane into the lumen. At the same time, splitting of water adds protons to the lumen, and reduction of NADPH removes protons from the stroma. The net result is a low pH in the thylakoid lumen, and a high pH in the stroma. ATP synthase uses this electrochemical gradient to make ATP. The majority of thylakoid lumen proteins with experimentally verified roles are involved in the function of either the PSII complex or the PSI–NDH supercomplex. The most abundant protein families in the thylakoid lumen are the OEC and OEC-like proteins (green), … The thylakoid lumen which is enclosed by the thylakoid membrane is a continuous aqueous phase. It is important especially for photophosphorylation in photosynthesis. The protons are pumped into the lumen via the membrane while reducing the pH level.

Alternatively, it is also hypothesized that CAH3 can remove a proton from the water-oxidizing complex on the donor The protein complexes involved in the photosynthetic light reactions are situated in the thylakoid membrane, which encloses a tiny space called lumen.

ATP synthase uses this electrochemical gradient to make ATP. Understanding the roles of the thylakoid lumen in photosynthesis regulation It has been known for a long time that the thylakoid lumen provides the environment for oxygen evolution, plastocyanin-mediated electron transfer, and photoprotection. the thylakoid membrane encloses a narrow, continuous com-partment, the lumen (1, 2).
